Your horseradish peroxidase (HRP) based ELISA can be more sensitive and specific through the use of our ColorBURST Blue substrate. ALerCHEK's single vial, liquid tetramethylbenzidine/peroxide formulation offers excellent stability and can detect as little as 0.0008pg of HRP, when terminated with 0.5N Sulfuric Acid and read at 450nm. ColorBURST Blue is as clear as water and is stable for at least 18 months at 4oC. Storage at 37oC for four weeks results in 90% of its original activity. It is sensitive to direct sunlight; however, it tolerates normal indoor light without auto oxidation. The reagent also tolerates inadvertent freeze thaw without loss of activity (a common problem during shipping).


ColorBURST Blue has extremely rapid kinetic properties which result in full color development for most assays within 20 minutes. Post termination stability is at least one hour without significant color fade.

Technology Behind ColorBURST Blue

3,5,3,'5' Tetramethylbenzidine (TMB) is not very water soluble and tends to precipitate easily. Additionally, the compound tends to auto-oxidize easily turning blue during formulation. This problem is further complicated by the fact that our formulation has both TMB, a potent reducing agent, and Peroxide, a potential oxidizing agent, in the same solution. Normally, this would be considered an incompatible situation.


After extensive formulations, ALerCHEK was able to bring these incompatible components together to form one ready to use reagent with optimized properties of sensitivity, specificity and stability all with a total absence of non-specific color.
